Could Citrus Peel Hold The Key to a Younger Liver?

It quietly juggles hundreds of jobs at once, breaking down nutrients, filtering nasty substances out of your blood, and somehow bouncing back from damage in a way most other organs simply can’t. It’s actually one of the only organs in the body capable of regenerating itself. But even the liver isn’t invincible, and as we get older, it starts to show signs of wear, becoming more prone to fatty liver disease and other age-related problems. New research suggests something rather ordinary: a compound found in citrus fruit, might actually help turn back the clock on an ageing liver.

The citrus compound behind the discovery

A team of researchers based in Taiwan has been studying a natural compound called hesperetin, found in lemons, limes, oranges, and other citrus fruits. It’s already known for a handful of promising properties, including reducing inflammation and potentially slowing down ageing processes in the body. What caught the researchers’ attention here was hesperetin’s link to a gene called CISD2, which plays a big role in keeping the liver and metabolism running smoothly.

According to the research team, when this gene isn’t active enough, it’s linked to fatty liver disease and even liver cancer. On the flip side, keeping the gene properly active seems to help preserve a more youthful liver and slow down age-related decline. The catch is that this gene naturally becomes less active as we get older, which is part of why livers tend to struggle more with age. That decline gave the researchers an idea, if hesperetin could boost activity of this gene, maybe it could help slow or even reverse some of that ageing process.

What happened when older mice were given the citrus compound

To test the theory, the team gave a group of naturally older mice, roughly equivalent to a senior citizen in human terms, a daily dose of hesperetin for five months. A separate group of mice the same age received an inactive substitute instead, purely as a comparison.

By the end of the five months, the untreated older mice showed the expected drop in that important gene’s activity. The mice given hesperetin, however, had activity levels that closely matched mice less than a quarter their age. On top of that, several other signs of an ageing liver improved too, including less fat buildup, fewer damaged liver cells, and reduced inflammation overall.

The researchers also looked closely at gene activity throughout the treated mice’s livers, and found that hesperetin had shifted things toward a noticeably younger-looking pattern, effectively undoing several of the changes typically seen with age. To double-check that this effect really was down to that specific gene, the team ran a separate experiment using mice bred without the gene at all in their liver cells.

Predictably, these mice already showed poor liver health at a young age, and giving them hesperetin made no meaningful difference at all, confirming that the gene is essential for the compound to actually work.

How the compound appears to work

The researchers believe hesperetin works through a chain reaction involving two other proteins in the body, which together help ramp up activity of that key longevity gene. It’s a fairly intricate biological pathway, but the basic takeaway is simple, this citrus compound appears to nudge the body into switching a protective gene back on.

Does this apply to humans too?

Naturally, the big question is whether any of this translates to people rather than just mice. To get a first look at that, the researchers examined liver tissue samples taken from 80 patients who’d previously had surgery related to liver tumours. They found that levels of the relevant proteins and genes declined with age in these human samples too, following a similar pattern to what was seen in the ageing mice. That doesn’t prove hesperetin would have the same rejuvenating effect in humans, but it does suggest the same biological process might be at play in us as well, which is a promising starting point for future research.

It’s still early days, and a lot more research will be needed before anyone can say with confidence that eating more citrus fruit, or taking a hesperetin supplement, could help protect your liver as you age. But the researchers behind this study are optimistic, describing their findings as a strong foundation for future work exploring hesperetin as a potential treatment to help slow liver ageing and reduce the risk of age-related fatty liver disease. Given everything your liver quietly does for you every single day, that’s a promising direction worth keeping an eye on.
